I believe that prior to the R-L Adapter came out, the only option was to stack adapters like you had suggested. I would think that the single R-L Adapter is the better solution,

but I would recommend reaching out to Dave Elwell at Leica in NJ for the best solution.

So just to clarify, with a novoflex adapter I wouldn't be able to manually select an R lens profile by going through the menu?

 

As a cheaper alternative, has anybody here tried a 200mm Nikkon AF D f/4 for macro work on the SL?  How does it compare to the 100 apo R?

Cheaper alternative if you don't mind manual focus … and which is usually better for life size macro imaging

…  the lens alone would be OK for 1:2 macro i.e. half life size.
 
Or this Leica R Macro Elmar 100/4 lens with bellows would be fine for 1:1 macro with an R to SL adapter 
 
 
… most of James Lager's Leica book illustrations were photographed with this lens; stopped down to f8 it's a super lens but not many photographers realise what a gem optic it is. 
 
Lighting and subject support and camera / lens support are just as important as the lens for close-up photography … and manual focus is preferred by many close-up enthusiasts.
 
Unlikely that an R lens profile (or any other 'profile') would be of much use (if any) for close-up imaging.
