I have no experience with video filming. 
Is there a fully automatic function or do you always have to adjust the exposure manually if the brightness changes during recording? 
That's how it works with the Q3.

Don't use CINE mode, that is all manual.

My recommendation would be to shoot in cine mode in order to statically set the variables. The last thing you want when shooting video is for ISO and white balance to change on you in the middle of a shoot. If you are unsure what to set statically, you can always turn everything on auto initially to find out what settings the camera thinks is best, then go into cine mode and set settings statically there using the values from Auto.

In addition, for video to look as filmic as possible as we are used to seeing in the movies, set shutter to 180°, shoot in 24p (if in the States, or shoot 25p if in Europe), shoot at 1/50, keep ISO as low as you can to avoid noise, and shot as wide open as you want, can, and need for DoF in your shot. 

In terms of focus, if you are just setting the camera up to film yourself, such as a talking head video, then yes put it on AFc. But if you are using it as a traditional cinematographer, you will likely want to pull focus. Turn on focus peaking, turn on manual focus, and wrack focus as needed.
